It's the central or overiding idea of the novel. There can certainly be secondary themes, as well. A common theme of many works is man's inhumanity to man. Great answers already... One more point to add to the discussion -- a common error is confusing a "topic" (one word) with a "theme" (complete thought). A theme really should be expressed as a complete sentence!
