Proper way to call back a job you applied for?

The manager said they would look over my application, and I've heard of people calling back the place they applied for and ask if they've looked over the resume. Whats the proper way to go about doing that/what are you supposed to say on the phone? (Note: This is a service job, food-industry)

Call back between 2:30-5 and ask for the manager. Don't say the step down thing.
