Should I ask to schedule an interview when doing a follow up call?
I applied for a job online last Saturday and if I do not get a call/email by this Saturday, I will call the hiring manager, check the status of my application, and express my interest in the position. If the position is still available,would it be okay if I ask the manager if it's possible to schedule an interview or should I just express my interest and tell the manager that I hope that he sees me as the best candidate for the job and hope that he will schedule an interview with me?

No frickin' way is that a good idea. If they wanted to schedule an interview with you they'd call you themselves. To assume that someone would take time out of their day to interview you just because you want one is as arrogant as it gets.
