There are so many places to get good cover letter resume templates online - I won't bore you with that here...
How do you feel about following up on a resume'?
BEFORE you submit your resume; find out if you know anyone working at this company - again. LinkedIn is a great resource. Then make sure when you submit resume you follow up with them and let them know. And yes, I always follow up with a quick email after I submit my resume.
What are your best tips for interviews?
Come prepared with a few questions. They are already interested in you, and your skill set matches the job description - which is why you got the interview. Don't assume you will get the job. Now is your chance to prove to them that you are the *best* person for the job.
